本研究在建立GMA诱导的HELF体外恶性转化模型的基础上 ,采用mRNA差异显示 (DD PCR)技术 ,对恶性转化细胞同正常细胞之间基因的差异表达状况进行了比较分析 ,共克隆到 18个差异表达cDNA片段。其中 ,17个DNA片段与已知基因高度同源 ,另外一个为新基因序列。Northern杂交证实 ,分别有 8个和 3个基因在GMA诱导的恶性转化细胞中表达增加或降低。结果提示 ,这些基因可能参与了GMA诱导的细胞恶性转化过程 ,为进一步研究GMA诱导细胞转化及其潜在致癌机理提供了新的线索
In this study, based on the GMA-induced malignant transformation of HELF in vitro model, differential display of mRNA (DD PCR) technology was used to analyze the differentially expressed genes in malignant transformed cells and normal cells. A total of 18 Differentially expressed cDNA fragments. Among them, 17 DNA fragments are highly homologous with known genes, and the other is a new gene sequence. Northern blotting confirmed that eight and three genes, respectively, increased or decreased in GMA-induced malignant transformed cells. The results suggest that these genes may be involved in the process of GMA-induced malignant transformation and provide new clues for further research on GMA-induced cell transformation and its potential carcinogenesis
