目的测定CHMFL-FLT3-122的2个解离常数。方法将CHMFL-FLT3-122溶于一系列的不同p H的缓冲溶液中,并在合适的波长处采用UV分别测定吸光度并计算其p Ka1值;以不同比例的乙腈-水(30∶70、40∶60、50∶50、60∶40)为溶剂配制不同浓度(0.009,0.013,0.017 mol·L~(-1))的CHMFL-FLT3-122溶液,用盐酸溶液滴定,利用非对数滴定法(Benet法)经数学计算推算出其p Ka2值。结果 CHMFL-FLT3-122存在2个解离常数,其p Ka1值为3.87±0.14,p Ka2值为8.12。结论采用UV和非对数滴定法对CHMFL-FLT3-122的解离常数进行测定,方法简单快速,准确可靠,可为其制剂的处方设计、质量标准的研究提供参考。
Objective To determine the two dissociation constants of CHMFL-FLT3-122. Methods CHMFL-FLT3-122 was dissolved in a series of buffer solutions of different p H, UV absorbance was measured at the appropriate wavelength and the p Ka1 value was calculated. Different proportions of acetonitrile-water (30:70, 40:60, 50:50 and 60:40) were used to prepare CHMFL-FLT3-122 solutions with different concentrations (0.009,0.013 and 0.017 mol·L -1) respectively, titrated with hydrochloric acid solution and non-logarithmic Method (Benet method) is calculated by mathematical calculation of its p Ka2 value. Results There were two dissociation constants for CHMFL-FLT3-122, with p Ka1 value of 3.87 ± 0.14 and p Ka2 value of 8.12. Conclusion The dissociation constant of CHMFL-FLT3-122 was determined by UV and non-logarithmic titration. The method was simple, rapid, accurate and reliable, and could provide a reference for the formulation design and quality standard of the preparation.
