目的通过黄芪等中药对大鼠子宫β-防御素-2基因表达的影响,探讨中药在局部组织提高机体免疫力的作用。方法将30只健康的Wistar大鼠随机分成空白组、黄芪组、苦参组、红芪组、黄芩组、双黄连组,分别灌胃,于第7日将大鼠处死并提取子宫组织RNA,用RT-PCR法检测大鼠β-防御素-2基因的表达变化。结果黄芪组、红芪组、双黄连组大鼠子宫组织β-防御素-2基因表达增强,而苦参组及黄芩组没有发生明显改变。结论黄芪、红芪、双黄连等中药可以增强大鼠子宫β-防御素-2基因表达,提示可以利用这些中药来增强局部组织的免疫力,以达到杀灭病原微生物的作用。
Objective To investigate the effects of Chinese herbal medicines such as Astragalus membranaceus on β-defensin-2 gene expression in rat uterus, and to explore the role of Chinese medicine in enhancing immunity of local organs. Methods Thirty healthy Wistar rats were randomly divided into blank group, Astragalus group, Sophora flavescens Astragali group, Radix Astragali group, Astragalus membranaceus group and Shuanghuanglian group. The rats were given intragastric administration. On the 7th day, rats were sacrificed and uterine tissue RNA was extracted. The expression of β-defensin-2 gene was detected by RT-PCR. Results The expression of β-defensin-2 gene was increased in the uterine tissue of the rats in the Astragalus membranaceus group, the Radix Scutellariae group, and the Shuanghuanglian group, but there was no significant change in the Sophora flavescens group and the Radix Astragali group. Conclusion Chinese herbs such as Huangqi, Hongqi and Shuanghuanglian can increase the expression of β-defensin-2 gene in rat uterus, suggesting that these herbs can be used to enhance the immunity of local tissues to achieve the role of killing pathogenic microorganisms.
