以[~3H]地塞米松为配体,应用Scatchard分析法测定30例糖尿病患者白细胞糖皮质激素受体(GR)的最大结合容量(Ro)及平衡解离常数(Kd)。结果表明Ⅰ型和Ⅱ型患者白细胞GR的Ro均明显降低,且亲和力也降低(Kd值增加)。应用Boyden小室法检测白细胞趋化移动(ChtM)及氢化可的松(F)对白细胞ChtM的抑制率(FI)。结果表明白细胞GR改变的同时伴有效应指标-F对白细胞ChtM的FI的相应变化。说明糖尿病患者白细胞GR的降低具有临床意义。
The maximal binding capacity (Ro) and equilibrium dissociation constant (Kd) of leukocyte glucocorticoid receptor (GR) in 30 diabetic patients were determined by Scatchard analysis using [~ 3H] dexamethasone as ligand. The results showed that type I and type II patients with leukocyte GR Ro significantly decreased, and the affinity is also reduced (Kd value increased). Boyden chamber method was used to detect leukocyte chemotaxis (ChtM) and hydrocortisone (F) inhibition rate of white blood cells (FI). The results showed that the change of leukocyte GR accompanied by the corresponding change of the effect index-F on the FI of leukocyte ChtM. Description of diabetic patients with leukocyte reduction of GR has clinical significance.
