目的:通过硫酸酯化修饰枸杞多糖,并对比研究其体内外抗肿瘤活性。方法:采用MTT法检测LBP_s对H_(22)肿瘤细胞的抑制率;建立H_(22)荷瘤小鼠模型,灌胃给药LBP_s,考察LBP_s对肿瘤细胞抑制率、对脾脏及胸腺的影响以及对血清中IL-2及TNF-α含量的影响。结果:LBP_s可显著抑制H_(22)肿瘤细胞增殖、提高免疫器官指数、提高血清中IL-2及TNF-α的含量,其活性优于未经修饰的LBP。结论:硫酸酯化修饰可显著提高枸杞多糖的抗肿瘤活性,该活性可能与免疫器官功能增强、IL-2及TNF-α释放增加有关。
OBJECTIVE: To purify lycium barbarum polysaccharides by sulfuric acid esterification and compare their anti-tumor activities in vitro and in vivo. Methods: The inhibitory rate of LBP_s on H_ (22) tumor cells was determined by MTT assay. H_ (22) tumor-bearing mouse model was established and intragastric administration of LBP_s was performed to investigate the effects of LBP_s on tumor cells, the spleen and thymus On serum IL-2 and TNF-α content. Results: LBP_s could significantly inhibit the proliferation of H22 cells, increase the index of immune organs and increase the levels of IL-2 and TNF-α in serum. The activity of LBP_s was better than that of unmodified LBP. CONCLUSION: Sulfated modification can significantly enhance the antitumor activity of LBP, which may be related to the enhancement of immune organs and the increase of IL-2 and TNF-α release.
