目的探讨科罗索酸对宫颈癌细胞Hela的凋亡诱导作用。方法不同浓度(10~50μmol·L~(-1))科罗索酸处理宫颈癌Hela细胞,MTT法检测细胞活性,SPSS12.0统计软件包计算半数抑制浓度(IC_(50)),流式细胞术检测细胞凋亡,荧光检测法检测凋亡酶caspase-3活性,western blot检测细胞蛋白Bax、Bcl-2表达。结果科罗索酸抑制宫颈癌Hela细胞活性随药物浓度及时间的增加而增高,24、48及72 h的IC_(50)分别为45、34、28μmol·L~(-1)。科罗索酸诱导Hela细胞凋亡率明显高于对照组(P<0.01),凋亡酶活性也明显高于对照组(P<0.01),科罗索酸可上调Bax而不影响Bcl-2的表达。结论科罗索酸可抑制宫颈癌Hela细胞活性,诱导细胞凋亡,其机制可能与上调Bax蛋白表达有关。
Objective To investigate the apoptosis-inducing effect of corosolic acid on cervical cancer Hela cells. Methods Cervical cancer Hela cells were treated with corosolic acid at different concentrations (10 ~ 50μmol·L ~ (-1)). The cell viability was measured by MTT assay. The half inhibitory concentration (IC 50) Cell apoptosis was detected by flow cytometry. Apoptotic activity of caspase-3 was detected by fluorescence assay. The expression of Bax and Bcl-2 was detected by western blot. Results Corosolic acid inhibited Hela cell activity of cervical cancer with the increase of drug concentration and time. The IC 50 of 24, 48 and 72 h were 45, 34 and 28 μmol·L -1, respectively. Corosolic acid induced apoptosis in Hela cells was significantly higher than that in control group (P <0.01), and apoptosis enzyme activity was also significantly higher than that in control group (P <0.01). Corosolic acid up-regulated Bax without affecting Bcl-2 expression. Conclusion Corosolic acid can inhibit Hela cell activity and induce apoptosis of cervical cancer cells. The mechanism may be related to the up-regulation of Bax protein expression.
