目的:观察不同浓度鹿茸提取液在正常和低钙条件下对离体蟾蜍心肌收缩力和心率的影响。方法:60只蟾蜍随机分为任氏液组对照组、任氏液+鹿茸组和低钙任氏液+鹿茸组,采用斯氏法制备离体蛙心灌流标本。在任氏液或低钙任氏液中加入鹿茸提取液,通过蟾蜍心室肌肉收缩波动,心室外加药心室内吸收的方法给药。应用BL-420F生物机能实验系统记录的离体蟾蜍心肌收缩曲线,并进行心肌收缩力和心率的统计学分析。结果:任氏液+鹿茸组:在心室外浓度达到40 mg·mL-1后,心肌收缩力在加药前后比较,心肌收缩力明显增加,P<0.01。低钙任氏液+鹿茸组:在心室外浓度达到5 mg·mL-1后,心肌收缩力在加药前后比较,心肌收缩力明显增加,P<0.05。正常蟾蜍离体蛙心灌流标本的心率、心室外加入鹿茸前后的心率和在低钙条件下心室外加入鹿茸前后的心率均没有显著变化,P>0.05。结论:在低钙条件下,鹿茸提取液对离体蟾蜍心脏活动的兴奋作用比在正常任氏液条件下更容易出现,但对心率没有影响。
Objective: To observe the effect of different concentrations of antler extract on contractile force and heart rate of isolated toad in normal and low calcium conditions. Methods: Sixty toads were randomly divided into two groups: control group, ringer’s solution + antler group and low calcium rinsus solution + antler group. Ringer’s solution or low calcium Ringer’s solution was added antler extract, by toad ventricular muscle contraction fluctuations, intraventricular administration of intraventricular absorption method of administration. The contractile curve of isolated toad myocardium recorded by the BL-420F bio-functional experimental system was used for statistical analysis of myocardial contractility and heart rate. Results: Ringer’s solution + antler group: The cardiac contractility was significantly increased before and after dosing at a concentration of 40 mg · mL-1 (P <0.01). Low calcium Ringer’s solution + antler group: The cardiac contractility increased significantly before and after dosing up to 5 mg · mL-1, P <0.05. There was no significant change in the heart rate of the toad isolated frog heart perfusion specimens, the heart rate before and after the addition of pilose antler, and the heart rate before and after the addition of velvet antler outside the heart under low calcium condition (P> 0.05). CONCLUSION: The antiepileptic effect of antler extract on isolated toad heart activity is more likely to occur at lower calcium concentrations than under normal Renre, but has no effect on heart rate.
