为探明不同色泽甜樱桃果实发育期间花色苷的积累及其与相关酶活性之间的关系,本研究以红色甜樱桃品种‘美早’和纯黄色品系‘13-33’为试材,对果实发育期间花色苷总量及相关酶活性进行了测定,并且采用超高效液相色谱-串联质谱技术(UPLC-MS/MS)对各花色苷组份进行了定性和定量分析。结果表明,从‘美早’果实中检测出7种花色苷,各组份含量伴随着果实的发育逐渐升高;从‘13-33’中仅检测出4种花色苷,各组份含量随果实的发育逐渐降低,到果实成熟期其含量已甚微。两个品种的花色苷总量与各组份含量的变化趋势相同;‘美早’中花色苷总量随着果实的发育逐渐升高,特别是在转色期后急剧升高;‘13-33’的花色苷总量随着果实的发育变化不明显,并且一直维持在极低水平。‘美早’的PAL、CHI、DFR和UFGT酶活性在果实发育后期均显著高于‘13-33’。相关性分析结果表明,‘美早’的花色苷总量与PAL和DFR酶活性之间无显著相关性,而与CHI和UFGT酶活性呈显著正相关。
In order to explore the relationship between the anthocyanin accumulation and the related enzyme activities during the development of sweet cherry fruit with different color, the present study was conducted with the red sweet cherry variety ’Mazao’ and the pure yellow ’13 -33 ’ Total anthocyanins and related enzyme activities during fruit development were determined, and each anthocyanin component was qualitatively and quantitatively analyzed using UPLC-MS / MS. The results showed that seven kinds of anthocyanins were detected from the fruits of ’Mei-Mei-Ao’, and the content of each component increased gradually with the development of fruit. Only four kinds of anthocyanins were detected from ’13 -33 ’ Fruit development gradually reduced to the fruit ripening stage its content has been minimal. The total anthocyanin content of the two cultivars was the same as that of each component. The total anthocyanin content of ’Mei-mei’was gradually increased with the development of fruit, especially after the color-changing period. ’13 - The total amount of anthocyanins in 33 ’was not obvious with the development of fruit, and remained at a very low level. The PAL, CHI, DFR and UFGT activities of ’Mihaicao’ were significantly higher than that of ’13 -33 ’during the later fruit development stage. The results of correlation analysis showed that there was no significant correlation between the total amount of anthocyanins and the activity of PAL and DFR, but significant positive correlation with the activities of CHI and UFGT.
