目的研究人乳头瘤病毒16型E6蛋白在COS-7细胞中的定位及表达。方法构建pEGFP-C1介导的HPV-16E6的真核表达载体pGFP-16E6,体外转染COS-7细胞,采用荧光显微镜观察HPV-16E6蛋白在COS-7细胞中的定位及表达。结果细胞在转染pGFP-16E6质粒后,细胞核发出明亮的绿色荧光,GFP-16E6于转染后6h开始表达,至21h表达到高峰,随后表达逐渐降低。结论HPV-16E6蛋白主要表达于COS-7细胞核内,并且表达随时间成动态变化。
Objective To study the localization and expression of human papillomavirus type 16 E6 protein in COS-7 cells. Methods The eukaryotic expression vector pGFP-16E6 of pEGFP-C1-mediated HPV-16E6 was constructed and transfected into COS-7 cells in vitro. The localization and expression of HPV-16E6 protein in COS-7 cells were observed by fluorescence microscopy. Results After transfection with pGFP-16E6 plasmid, the nuclei of cells showed bright green fluorescence. GFP-16E6 began to express at 6h after transfection and peaked at 21h, then decreased gradually. Conclusion HPV-16E6 protein is mainly expressed in the nucleus of COS-7 cells, and its expression changes dynamically with time.
