本文报道用国产高效液相色谱仪联电化学检测器,反相内标法测定血清去甲肾上腺素(NA)及肾上腺素(AD)。研究了不同流动相条件下对血清NA及AD容量因子(K′)及响应值的影响,从而选择最佳测定条件。血清NA及AD浓度在0.2~10ng/ml内与色谱峰面积比值的线性关系良好(r>0.99);不同浓度的血清NA和AD绝对回收率及方法回收率分别为74.4~89.9%及95.1~107.6%;不同浓度的日内、日间变异系数分别为5.0~9.7%及5.3~11.9%。测定10名正常人及4例嗜铬细胞瘤患者的术前血清NA和AD值,二者有显著差异。提示本法对诊断嗜铬细胞瘤有一定的临床价值。
This article reports the use of domestic high performance liquid chromatography coupled electrochemical detector, reverse phase internal standard method for the determination of serum norepinephrine (NA) and adrenaline (AD). The influence of different mobile phase conditions on serum NA and AD capacity factor (K ’) and response value was studied to select the best assay conditions. The linear relationship between serum NA and AD concentration and peak area ratio was good (r> 0.99) in the range of 0.2 ~ 10 ng / ml. The absolute recoveries and recoveries of serum NA and AD were 74.4-89.9% and 95.1 ~ 107.6%. The intra-day and inter-day variation coefficients of different concentrations were 5.0-9.7% and 5.3-11.9% respectively. Preoperative serum NA and AD values were determined in 10 normal and 4 pheochromocytoma patients, both of which were significantly different. Prompted this law on the diagnosis of pheochromocytoma have some clinical value.
