目的 研究脑胶质瘤中IL 6 /IL 6R自分泌或旁分泌环路的存在和意义。方法 采用逆转录聚合酶链反应 (RT PCR)方法检测白细胞介素 6 (IL 6 )和IL 6受体 (IL 6R)基因在脑胶质瘤细胞株、脑胶质瘤组织标本和正常人脑胶质细胞中的表达。结果 2株脑胶质瘤细胞株均有IL 6和IL 6R的表达 ,5 2例脑胶质瘤组织标本中有 38例 (73.1% )表达IL 6、4 4例 (84 .6 % )表达IL 6R、33例 (6 3.5 % )同时表达IL 6和IL 6R。人脑正常胶质细胞仅有IL 6基因弱表达 ,而无IL 6R表达。结论 脑胶质瘤可能存在IL 6 /IL 6R自分泌或旁分泌环路 ,此环路与肿瘤的恶性增殖有关。
Objective To study the existence and significance of the autocrine or paracrine loop of IL 6/IL 6R in gliomas. Methods Reverse transcription polymerase chain reaction (RT PCR) was used to detect interleukin 6 (IL 6) and IL 6 receptor (IL 6R) genes in glioma cell lines, glioma tissues, and normal human brains. Expression in glial cells. Results The expressions of IL 6 and IL 6R were detected in 2 glioma cell lines. Of the 52 glioma tissue samples, 38 (73.1%) expressed IL 6 and 44 (84.6%) were expressed. IL 6R and 33 cases (6 3.5 %) expressed IL 6 and IL 6R simultaneously. Only IL 6 gene was weakly expressed in normal human brain glial cells, but no IL 6R was expressed. Conclusion There may be an autocrine or paracrine loop of IL 6/IL 6R in glioma, which is related to the malignant proliferation of tumors.
