目的研究四川省问号钩端螺旋体(简称钩体)中毒力相关基因分布特点,为防治钩体病提供科学依据。方法根据钩体赖型赖株全基因组序列生物信息学资料,选择7个钩体致病相关基因与群特异性G1、G2基因,采用聚合酶链反应(PCR),对四川省1970-2010年分离保存的问号钩体90株和对照用双曲钩体4株,共94株进行了检测。结果 90株问号钩体中7个毒力相关基因,除sphA基因检出率较低外(6.67%),其余6个毒力相关基因检出率在81.1%~98.9%之间,G1、G2基因检出率为100%,双曲钩体中未检测出7个毒力相关基因和G1、G2基因。结论四川省问号钩体中毒力相关基因分布广泛,检出率高,患者分离株与黑线姬鼠分离株毒力相关基因分布高度一致。
Objective To study the distribution characteristics of virulence-related genes in Leptospira interrogans in Sichuan province (referred to as leptospira), and provide a scientific basis for the prevention and control of leptospirosis. Methods According to the bioinformatics data of Leptospira interrogans strains, seven pathogen-related genes and group-specific G1 and G2 genes were selected and sequenced by PCR. Ninety-four strains of Leptospira interrogans and four strains of Leptospira interrogans were isolated and identified, and a total of 94 strains were tested. Results The detection rate of 7 virulence genes in 90 ticks was lower than that of sphA gene (6.67%), and the other 6 virulence genes were between 81.1% and 98.9% The gene detection rate was 100%. No seven virulence-related genes and G1, G2 genes were detected in the leptospira. Conclusions The virulence-associated genes of L. aeruginosa were widely distributed and detected in Sichuan Province. The distribution of virulence-related genes in the isolates of Apodemus agrarius from patients was highly consistent.
