目的研究大鼠肝组织卵圆细胞的定位及其超微结构。方法建立SD大鼠卵圆细胞增生模型,用胆管上皮分化标志CK18、19和干细胞标志CD34作组织免疫组化染色,采用透射电镜观察超微结构。结果组织免疫组化发现大鼠卵圆细胞主要分布于汇管区,部分分散于肝小叶内,根据组织和细胞透射电镜超微结构特点,发现卵圆细胞有三型,Ⅰ型细胞体积较小、7μm左右,核大、胞质少,细胞器少,此为较为原始的卵圆细胞。Ⅱ型细胞体积稍大,8μm左右,胞质稍多,有部分细胞器。Ⅲ型细胞体积更大,9μm左右,细胞器较多。结论大鼠肝卵圆细胞位于汇管区,超微结构观察发现肝卵圆细胞可能是肝脏干细胞。
Objective To study the localization and ultrastructure of oval cells in rat liver tissue. Methods The model of oval cell proliferation in SD rats was established. Immunohistochemical staining of bile duct epithelial differentiation markers CK18, 19 and stem cell marker CD34 was used to observe the ultrastructure. Results Immunohistochemistry showed that the oval cells of rat were mainly distributed in the portal area and partially dispersed in the hepatic lobule. According to the ultrastructure of tissue and cell TEM, three types of oval cells were found, and the type I cells were smaller and 7μm Or so, nuclear large, less cytoplasm, organelles less, this is the more primitive oval cells. Type Ⅱ cells slightly larger, about 8μm, slightly more cytoplasm, some organelles. Type III cells larger, about 9μm, organelles more. Conclusions The rat hepatic oval cells are located in the portal area. The ultrastructure observation shows that hepatic oval cells may be hepatic stem cells.
