目的:探讨电针联合脂肪源性干细胞(ADSC)移植对大鼠缺血/再灌注损伤后海马区凋亡相关基因Livin和Caspase-3表达的影响。方法:采用线栓法复制大鼠大脑中动脉缺血再灌注模型,将动物随机分为模型组、电针组、ADSC移植组、电针+ADSC组。应用NSS法行神经功能损害评分,采用蛋白质印迹法检测海马区Livin和Caspase-3蛋白表达水平。结果:电针+ADSC组海马区PKH-26标记的细胞个数高于ADSC组(P<0.05)。电针+ADSC组NSS评分低于单独治疗组(P<0.05)。与模型组比较,电针+ADSC组和电针组Livin蛋白表达增高(P<0.05),Caspase-3蛋白表达下降(P<0.05)。结论:电针联合脂肪源性干细胞移植促进脑缺血大鼠神经功能恢复作用优于单独治疗组,其机制可能与电针通过上调海马区Livin表达,下调Caspase-3表达,抑制移植的ADSC凋亡有关。
Objective: To investigate the effects of electroacupuncture combined with adipose derived stem cells (ADSCs) on the expression of Livin and Caspase-3 in hippocampus after ischemia / reperfusion injury in rats. Methods: The rat middle cerebral artery occlusion (MCAO) model was reproduced by thread occlusion. Animals were randomly divided into model group, EA group, ADSC transplantation group and electroacupuncture + ADSC group. Nerve function impairment score was determined by NSS method. The protein expression of Livin and Caspase-3 in hippocampus was detected by Western blotting. Results: The number of PKH-26-labeled cells in hippocampus of EA + ADSC group was higher than that in ADSC group (P <0.05). The electroacupuncture + ADSC group NSS score was lower than the single treatment group (P <0.05). Compared with the model group, the expression of Livin protein increased (P <0.05) and the expression of Caspase-3 protein decreased in EA group + ADSC group and EA group (P <0.05). CONCLUSION: Electroacupuncture combined with adipose-derived stem cell transplantation can promote the neurological function recovery of rats with focal cerebral ischemia and is better than that of the untreated group. The mechanism may be related to the fact that electroacupuncture can reduce the expression of Livin, downregulate the expression of Caspase-3, Death related.
