高迁移率族蛋白B-1(high mobility group box-B1,HMGB1),是一种非组蛋白染色体蛋白质,在正常情况下位于细胞内,参与多种生物学过程,包括维持核小体结构、调节基因转录、核糖核酸形成、DNA修复、V(D)J重组、分化和发展等。HMGB1主要参与炎症反应,并与肿瘤细胞的生长、浸润和转移有密切关系。HMGB1可能是一种抗凋亡结合蛋白,其过表达可抑制细胞凋亡,从而导致肿瘤的发生、生长。另外,HMGB1与纤维蛋白溶酶催化剂系统、基质金属蛋白酶的活化和促进黏附细胞迁移有关,这些与细胞侵袭、转移有关。拮抗HMGB1能起到抑制肿瘤生长、浸润及转移的作用。
High mobility group box-B1 (HMGB1), a non-histone chromosomal protein, is normally located in the cell and participates in a variety of biological processes including the maintenance of the nucleosome structure, Regulate gene transcription, RNA formation, DNA repair, V (D) J recombination, differentiation and development. HMGB1 is mainly involved in inflammatory reaction, which is closely related to tumor cell growth, invasion and metastasis. HMGB1 may be an anti-apoptotic binding protein whose overexpression can inhibit apoptosis, leading to tumorigenesis and growth. In addition, HMGB1 is associated with activation of the plasmin catalyst system, matrix metalloproteinase, and adhesion cell migration, which are involved in cell invasion and metastasis. Antagonizing HMGB1 can play a role in inhibiting tumor growth, invasion and metastasis.
