目的 :研究氢氟酸烧伤中毒对兔外周血单个核细胞 (PBMC)早期凋亡百分率和胞内游离钙浓度([Ca2 + ]i)的影响。方法 :用流式细胞仪检测兔烧伤染毒前后外周血单个核细胞的AnnexinV变化 ,以观察其早期凋亡百分率。和用Fluo - 3/Am荧光探针观察烧伤染毒前后外周血单个核细胞内Ca2 + 平均荧光强度值的变化 ,以观察细胞内 [Ca2 + ]i的变化。结果 :12只烧伤中毒兔外周血单个核细胞早期凋亡百分率显著增加 ,烧伤染毒前后比较P<0 0 1。而 12只烧伤中毒兔中 8只染毒后 1h外周血单个核细胞内 [Ca2 + ]i显著降低 ,烧伤染毒前后比较P <0 0 5。其余 4只却表现 [Ca2 + ]i染毒前后比较P >0 0 5。结论 :本实验氢氟酸烧伤中毒使兔外周血单个核细胞早期凋亡百分率显著增加 ,外周血单个核细胞内 [Ca2 + ]i却显著降低。提示氢氟酸烧伤中毒引导的细胞凋亡并非细胞内 [Ca2 + ]i增加所引发
Objective: To study the effect of hydrofluoric acid burn on the percentage of early apoptosis and intracellular free calcium concentration ([Ca2 +] i) in peripheral blood mononuclear cells (PBMCs) of rabbits. Methods: The changes of Annexin V in peripheral blood mononuclear cells before and after burn treatment were detected by flow cytometry to observe the percentage of early apoptosis. Fluorescence probes of Fluo - 3 / Am were used to observe the change of intracellular Ca2 + fluorescence intensity in peripheral blood mononuclear cells before and after burn treatment in order to observe the change of intracellular [Ca2 +] i. Results: The percentages of early apoptosis of peripheral blood mononuclear cells in 12 rabbits were significantly increased compared with those before burn and post burn (P <0.01). However, the level of [Ca2 +] i in peripheral blood mononuclear cells was significantly decreased at 1 hour after exposure to 8 burn poisoning rabbits in 12 burn poisoning groups, P <0 05 before and after burn injury. The remaining four showed [Ca2 +] i compared before and after P> 0 0 5. CONCLUSION: Hydrofluoric acid burn poisoning can significantly increase the early apoptotic percentage of peripheral blood mononuclear cells in peripheral blood mononuclear cells, and decrease the [Ca2 +] i in peripheral blood mononuclear cells.
