目的利用传统的检验方法与实时荧光PCR技术相结合检测军团菌。方法按照卫生部《公共场所集中空调通风系统卫生规范附录A》(2006)的要求,于2010年10月—2011年12月,对深圳市6家宾馆酒店86件淋浴热水进行军团菌检测。采用传统细菌分离培养法进行分离鉴定,采用实时荧光PCR法加以验证。结果在86件淋浴热水中,20件检出嗜肺军团菌,嗜肺军团菌阳性率为23.3%,其中,Lp1、Lp3、Lp6血清型分别占65%,10%,25%;实时荧光PCR法鉴定出1件淋浴热水为非嗜肺的军团菌属。结论深圳市宾馆酒店淋浴热水存在军团菌的污染;传统的检验方法与荧光PCR技术的检测结果具有较高的一致性。
Objective To detect Legionella by combining traditional test methods with real-time fluorescence PCR. Methods According to the requirements of Ministry of Health, “Hygienic Regulations of Central Air Conditioning Ventilation System in Public Places” (Appendix A), from October 2010 to December 2011, 86 Legionella samples were tested for hot water in six guesthouses in Shenzhen. Isolation and identification using traditional bacterial isolation and culture methods, using real-time fluorescent PCR method to be verified. Results The positive rate of Legionella pneumophila and Legionella pneumophila in 20 out of 86 showering hot water samples was 23.3%, of which Lp1, Lp3 and Lp6 serotypes accounted for 65%, 10% and 25%, respectively. Real-time fluorescence PCR method identified a shower hot water non Legionella genus Legionella. Conclusion There is Legionella contamination in the hot shower of hotel in Shenzhen City. The traditional test methods are in good agreement with the test results of fluorescent PCR.
