从山东肥城采集到香菇病害样品,分离得到病原菌分离物FC,提取其基因组总DNA,利用真菌通用引物ITS1/ITS4扩增其r DNA ITS区域序列并进行测序。将获得的序列进行同源性比对和构建系统进化树,发现FC分离物与Gen Bank中8个米根霉(R.oryzae)分离物同源性最高,为98.27%,结合形态学特征,证明该分离物为米根霉(R.oryzae)。
The samples of lentinula edodes were collected from Feicheng, Shandong province. FC of pathogen was isolated and the total genomic DNA was extracted. ITS region of rDNA was amplified by ITS1 / ITS4 and sequenced. The obtained sequences were aligned by homology and phylogenetic tree was constructed. It was found that FC isolates had the highest homology with eight R. oryzae isolates in Gen Bank (98.27%). According to morphological characteristics, The isolate proved to be R. oryzae.
