目的 :研究苦参碱的利尿作用及作用时间与其药代动力学特征之间的相关性。方法 :运用家兔导尿管集尿法 ,观察灌服苦参碱后家兔尿量的变化。用HPLC法测定苦参碱在大鼠口服后的血浆浓度。结果 :苦参碱 90mg/kg灌服可显著增加家兔的尿量 (P <0 .0 1)。最大效应出现在灌服的 60min后 ,且维持时间可达灌服后的 180min ,大鼠口服后血浆药物达峰时间Tmax平均为 55min ,t1/ 2 β平均为10 2min。结论 :苦参碱对家兔有显著的利尿作用 ,其效应的维持相对滞后于药物在血浆中的衰减。
Objective: To study the relationship between the diuretic effect and the duration of action of matrine and its pharmacokinetic characteristics. Methods :Using rabbit catheter collection method to observe the change of urine volume after rabbits fed matrine. The plasma concentration of matrine after oral administration in rats was determined by HPLC. RESULTS: Matrine 90mg/kg orally could significantly increase the urine output of rabbits (P < 0.01). The maximal effect occurred after 60 minutes of oral administration, and the maintenance time was 180 minutes after oral administration. The average Tmax of plasma drug peak time after oral administration in rats was 55 min, and the average t 1 / 2 β was 102 min. Conclusion : Matrine has a significant diuretic effect on rabbits, and the maintenance of its effect lags behind that of drugs in plasma.
