目的 研究转化生长因子 α(transforminggrowthfactor α ,TGF α)在胚胎植入早期阶段的作用。方法 小鼠胚泡体外培养于含或不含各种浓度的TGF α培养液中。用粘附、扩展胚胎的百分数来评价胚泡的着床能力。用明胶酶谱法来检测胚泡分泌基质金属蛋白酶情况。结果 实验组与对照组的胚泡粘附率无明显差异 ,但是培养 2 4h后 ,实验组胚泡扩展率显著高于对照组。明胶酶谱结果显示实验组胚泡分泌基质金属蛋白酶 (matrixmetalloproteinase ,MMPs)早于对照组。结论 TGF α通过促进胚泡扩展和基质金属蛋白酶分泌参与到胚泡植入进程的调节当中。
Objective To investigate the role of transforming growth factor α (TGF α) in the early stage of embryo implantation. Methods Mouse blastocysts were cultured in vitro with or without TGFβ in various concentrations. The rate of blastocyst implantation was evaluated by the percentage of adherent, expanded embryos. Gelatin zymography was used to detect the status of blastocyst secretion of matrix metalloproteinases. Results There was no significant difference in blastocyst adhesion between the experimental group and the control group, but the blastocyst expansion rate of the experimental group was significantly higher than that of the control group after culturing for 24 hours. Gelatin zymography showed that the blastocyst in the experimental group secreted matrix metalloproteinase (MMPs) earlier than the control group. Conclusion TGF α is involved in the regulation of blastocyst implantation by promoting blastocyst expansion and matrix metalloproteinase secretion.
