[Objective] To screen and optimize the extracting technology for GANODERMA capsule. [Methods]Lymphocyte transformation test in mice induced by Con A was used to screen the three extracting technologies,which were ethanol-extracting technology,water-ethanol extracting technology and water-extracting technology. With extraction rate and total amount of GANODERMA polysaccharides as the investigation indices,orthogonal test was designed. Effects of water volume,extraction temperature and extraction time on the extraction technology of GANODERMA were investigated. [Results]Results of animal test showed that water-extracting technology had the optimal effects of immunoregulation on mice. And the optimal water-extracting technology of GANODERMA capsule was as follows: after two times of boiling for GANODERMA at 100 ℃,the first extraction was carried out according to 1∶ 20 solid- liquid ratio(g /mL); then reflux extraction was carried out for 2 h after boiling; after filtration,1∶ 15 solid- liquid ratio(g /mL) was carried out for the second extraction,then reflux extraction was adopted for2 h after boiling; the second extraction was carried out according to 1∶ 15 solid-liquid ratio(g /mL),then reflux extraction was adopted for1 h after boiling. [Conclusions]Water-extracting technology for GANODERMA capsule had the optimal effects of immunoregulation; and the selected extraction technology was reasonable and feasible.
