目的探讨葡萄糖调节蛋白78(GRP78)mRNA及蛋白在肝癌和癌旁肝硬化组织中的表达情况及其与肝癌发生的关系。方法肝癌手术切除标本30份,应用半定量逆转录—聚合酶链反应(RT-PCR)分别检测肝癌组织和癌旁肝硬化组织中GRP78mRNA的相对表达量,并通过免疫组化SP法检测肝癌组织和癌旁肝硬化组织中GRP78蛋白的表达情况。结果①半定量RT-PCR显示,GRP78mRNA在所有肝癌组织中均呈高表达,相对表达量为1.7037±0.8501,而在癌旁肝硬化组织中呈低表达或不表达,相对表达量为0.2742±0.3452,2组之间差异具有统计学意义(P<0.01)。②免疫组织化学及图像分析结果显示,在肝癌组织和癌旁肝硬化组织中GRP78蛋白平均灰度扫描值分别为171.22±20.9705及139.53±20.2320,2组之间亦存在统计学意义(P<0.01)。结论GRP78在肝癌中的表达明显增高,可能在肝癌的发生、发展过程中具有重要意义。
Objective To investigate the expression of glucose regulatory protein 78 (GRP78) mRNA and protein in hepatocellular carcinoma and para-cancerous liver cirrhosis and its relationship with the development of hepatocellular carcinoma. Methods The expression of GRP78 mRNA in hepatocellular carcinoma tissues and adjacent cirrhosis tissues was detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). The expression of GRP78 mRNA in 30 specimens of hepatocellular carcinoma was detected by immunohistochemical SP method. And adjacent liver cirrhosis GRP78 protein expression. Results ① Semi-quantitative RT-PCR showed that GRP78 mRNA was highly expressed in all hepatocellular carcinoma tissues with a relative expression level of 1.7037 ± 0.8501 and a low or no expression in paracancerous cirrhotic tissues with a relative expression level of 0.2742 ± 0.3452 , The difference between the two groups was statistically significant (P <0.01). ② The results of immunohistochemistry and image analysis showed that the mean grayscale gray value of GRP78 protein in HCC tissues and adjacent liver cirrhosis tissues were 171.22 ± 20.9705 and 139.53 ± 20.2320, respectively (P <0.01 ). Conclusion The expression of GRP78 in hepatocellular carcinoma was significantly increased, which may be of great significance in the occurrence and development of hepatocellular carcinoma.
