目的探讨使用反相高效液相色谱法测定他克莫司血药浓度的方法。方法采用色谱柱Diamonsil C18柱(250 mm×4.6 mm,5μm),流动相为乙腈∶0.2%磷酸溶液∶四氢呋喃(55∶40∶5),流速1.0 ml/min,柱温30℃,检测波长210 nm。结果他克莫司血药浓度在1.3~15.6μg/ml(r=0.9996,n=6)范围内线性关系良好,平均加样回收率为101.4%。结论本法简便易行,适用于他克莫司血药浓度测定和药动学研究。
Objective To investigate the method of determining the concentration of tacrolimus in blood by reversed-phase high performance liquid chromatography. Methods Diamonsil C18 column (250 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase was acetonitrile: 0.2% phosphoric acid solution: tetrahydrofuran (55:40:5), the flow rate was 1.0 ml / nm. Results The plasma concentration of tacrolimus was linear within the range of 1.3 ~ 15.6μg / ml (r = 0.9996, n = 6) with an average recovery of 101.4%. Conclusion This method is simple and convenient, suitable for the determination of tacrolimus blood concentration and pharmacokinetics.
