目的:建立耐药性骨肉瘤小鼠模型,并确定其生物学特性。方法:通过建立的阿霉素耐药性骨肉瘤细胞KH-OS,将其移植到SCID小鼠中,观察小鼠生物学特性,包括荷瘤大小、病理学以及耐药基因的免疫组化检测。结果:2周内小鼠体内明显成瘤,病理学检测提示为骨细胞瘤,耐药相关基因MDR1、GST-π和Topo-Ⅱ在KH-OS/ADM移植的骨肉瘤小鼠模型实验组中50%表达。结论:通过耐药性骨肉瘤细胞系可建立耐药性的骨肉瘤小鼠模型,可用于骨肉瘤动物体内耐药性机制的科研实验。
Objective: To establish a mouse model of drug-resistant osteosarcoma and determine its biological characteristics. Methods: Adriamycin-resistant osteosarcoma KH-OS cells were transplanted into SCID mice to observe the biological characteristics of the mice, including tumor size, pathology and immunohistochemical detection of drug resistance genes . Results: The mice developed tumorigenicity within 2 weeks. The pathological examination indicated that the expression of multidrug resistance-related genes MDR1, GST-π and Topo-Ⅱ were significantly increased in KH-OS / ADM transplanted osteosarcoma mice 50% expression. Conclusion: The drug-resistant osteosarcoma mouse model can be established by drug-resistant osteosarcoma cell line and can be used in scientific research of drug resistance mechanism in osteosarcoma animal.
