目的建立地骨皮中甜菜碱的高效液相色谱-蒸发光散射检测器分析方法。方法固定相为Luna C18色谱柱(250 mm×4 mm,5μm),柱温30℃;水-冰醋酸(99.9∶0.1)为流动相,流速0.15 mL/min;蒸发光散射检测器检测,漂移管温度100℃,气体流速2.50 L/min。结果甜菜碱在0.486~2.430μg范围内线性关系良好,相关系数r为0.9999;地骨皮药材中甜菜碱的平均回收率为97.4%,RSD为1.3%(n=5)。结论该方法准确,重现性好,可作为地骨皮质量控制的方法。
Objective To establish a high performance liquid chromatography-evaporative light scattering detector analysis method for betaine in ground bony skin. Methods The stationary phase was Luna C18 column (250 mm×4 mm, 5μm), column temperature 30°C; water-glacial acetic acid (99.9∶0.1) as mobile phase, flow rate 0.15 mL/min; detection by evaporative light scattering detector, drift The tube temperature is 100°C and the gas flow rate is 2.50 L/min. The results showed that the linear relationship of betaine was good in the range of 0.486~2.430μg, and the correlation coefficient r was 0.9999. The average recovery rate of betaine was 97.4% and the RSD was 1.3% (n=5). Conclusion This method is accurate and has good reproducibility. It can be used as a method to control the quality of ground bony skin.
