应用RAPD标记对原产于云南等地的 2 4份野生茶树资源进行分子鉴定研究。结果表明 ,RAPD标记在鉴定茶树种质资源方面非常有效。有 3种独立的方法可以用于茶树种质资源的分子鉴定 :特殊的标记 ;特异的谱带类型 ;不同引物提供谱带类型的组合。 16个特异标记的存在和 3个特异标记的缺失可以鉴定 14份资源 ;OPO 13扩增的 13种谱带类型可以鉴定 10份资源。利用最少数量引物获得最大鉴定能力 ,对种质资源的分子鉴定尤为重要。OPO 13、OPO 18、OPG 12和OPA 13等 4个引物带型的组合则可以鉴定所有 2 4份资源 ,包括形态和生化成分上几乎没有差异的 2株毗邻野生茶树
RAPD markers were used to identify 24 wild tea plants originated in Yunnan and other places. The results showed that the RAPD marker was very effective in identifying tea germplasm resources. Three independent methods are available for the molecular identification of tea germplasm: special markers; specific band types; and different primers provide a combination of band types. The presence of 16 specific markers and the deletion of 3 specific markers allowed the identification of 14 resources; the 13 bands amplified by OPO 13 identified 10 resources. The use of a minimum number of primers to obtain the maximum identification ability, molecular identification of germplasm resources is particularly important. The combination of four primer bands, OPO 13, OPO 18, OPG 12 and OPA 13, identified all 24 resources including 2 closely-related wild tea trees with almost no difference in morphological and biochemical composition
