目的探索二至丸有效成分对损伤后肝细胞再生障碍大鼠肝细胞凋亡水平的影响。方法将40只Wistar大鼠随机分为4组(即正常组、模型组、齐墩果酸+红景天苷组、雷帕霉素组),除正常组建立肝部分切除术(partial hepatectomy,PHx)模型外,其余各组建立损伤后肝细胞再生抑制复合模型(PHx+2AAF),齐墩果酸+红景天苷治疗性给药3d后处死。分离肝脏,计算肝重指数,制作病理切片进行形态学观察,并采用自动生化分析仪检测血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、γ-谷酰转肽酶(γ-GT)活性、血清碱性磷酸酶(AKP)、白蛋白、总胆红素(TBIL)、直接胆红素(DBIL)水平。结果与模型组比较,齐墩果酸+红景天苷组大鼠肝重、肝重指数、白蛋白及DBIL水平显著升高(P<0.05或P<0.01),而AKP、γ-GT活性、TBIL、AST、ALT则明显下降(P<0.05或P<0.01),同时病理形态学表明,给药后肝细胞排列紧密,中央静脉正常、或充血,少量炎细胞浸润,少量肝细胞脂肪变性,提示大鼠肝脏病理损伤明显缓解。结论二至丸有效成分对大鼠损伤后肝细胞再生障碍具有较明显改善作用。
Objective To explore the effect of active ingredients of Erzhi Pills on hepatocyte apoptosis in rats with hepatocellular aplasia after injury. Methods Forty Wistar rats were randomly divided into four groups (normal group, model group, oleanolic acid + salidroside group, rapamycin group). In addition to the normal hepatic resection (partial hepatectomy, PHx) model, the rest of the groups to establish a composite model of post-injury hepatocyte regeneration inhibition (PHx +2 2AAF), oleanolic acid + salidroside therapeutic 3d after sacrifice. The liver was separated and the liver index was calculated. Pathological sections were made for morphological observation. Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), γ-glutamyltransferase Peptidase (γ-GT) activity, serum alkaline phosphatase (AKP), albumin, total bilirubin (TBIL), direct bilirubin (DBIL) Results Compared with the model group, the liver weight, liver index, albumin and DBIL in the oleanolic acid + salidroside group were significantly increased (P <0.05 or P <0.01), while the activity of AKP and γ-GT , TBIL, AST and ALT decreased significantly (P <0.05 or P <0.01). At the same time, the histopathology showed that the hepatocytes arranged closely, the central veins were normal, or hyperemia, infiltration of inflammatory cells and a small amount of steatosis , Suggesting that rat liver pathological injury significantly alleviated. Conclusion The active ingredient of Erzhi Pills can significantly improve the disorder of hepatocyte regeneration after injury in rats.
