目的研究凹顶藻萜类化合物(Laurencia terpenoid extract,LET)对接种H_(22)瘤细胞小鼠白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平的影响。方法1.以反相高效液相色谱法测定LET中总萜的含量;2.昆明小鼠随机分为5组(10只/组),即模型组、LET低中高剂量组(25,50,100mg·kg~(-1)·d~(-1))、环磷酰胺组(CY对照组)。建立小鼠肝癌H_(22)移植性肿瘤模型,次日除模型组外,其余4组分别以不同剂量的LET、CY灌胃,共15d后处死,准确称取瘤重,计算抑瘤率,并采用放免法(RIA)测定血清中IL-6、TNF-α的水平。结果LET灌胃给药小鼠H_(22)肿瘤质量增长较模型组缓慢,抑瘤率分别为27.5%、34.9%和41.4%;且能升高细胞因子IL-6、TNF-α的水平。结论LET能较好地抑制H_(22)小鼠肿瘤增长,其抗肿瘤机制可能与升高机体细胞因子IL-6、TNF-α水平有关。
Objective To investigate the effects of Laurencia terpenoid extract (LET) on the levels of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in mice inoculated with H_22 tumor cells. Method 1. Determination of total quinone in LET by reversed-phase high performance liquid chromatography; 2. Kunming mice were randomly divided into 5 groups (10/group), namely model group, LET low-medium high-dose group (25, 50, 100mg ·kg~(-1)·d~(-1)), cyclophosphamide group (CY control group). A mouse H_(22) transplantable tumor model was established. On the next day except for the model group, the other 4 groups were given different doses of LET and CY, respectively, and were sacrificed 15 days later. The tumor weight was accurately weighed and the tumor inhibition rate was calculated. Radioimmunoassay (RIA) was used to measure serum IL-6 and TNF-α levels. Results The tumor growth rate of H_(22) mice fed with LET was slower than that of the model group. The tumor inhibition rates were 27.5%, 34.9% and 41.4%, respectively; and the levels of cytokines IL-6 and TNF-α were increased. Conclusion LET can inhibit the tumor growth of H22 mice, and its anti-tumor mechanism may be related to the increase of cytokine levels of IL-6 and TNF-α.
