目的:研究Survivin基因在大肠癌中的表达及其与肿瘤发生、发展的关系。方法:流式细胞术(FlowCytometry,FCM)检测58例大肠癌(Colorectal carcinoma,CC)患者及10例正常者黏膜(Normal mucosa,NM)中Survivin表达情况,并结合临床病理相关因素,应用SPSS11.0统计软件进行统计学分析。结果:FCM定量检测表明,正常黏膜组Survivin表达平均FI值为1.00±0.28,高、中、低分化腺癌及粘液腺癌组Survivin表达FI值分别为3.16±1.39、3.20±1.04、3.32±0.48和3.31±1.06。高、中、低分化腺癌及粘液腺癌组Survivin表达均明显高于正常黏膜组(P<0.05)。大肠癌淋巴结转移组平均FI值3.11±1.19,显著高于无淋巴结转移组2.26±1.38(P<0.05)。FCM结果显示Survivin表达与患者的年龄、性别、肿瘤的发生部位等无关(P>0.05)。结论:Survivin在大肠癌中表达异常增高,显著高于正常肠黏膜组织。Survivin在大肠癌中的表达与淋巴结转移密切相关。
Objective: To investigate the expression of survivin gene in colorectal carcinoma and its relationship with tumorigenesis and development. Methods: Flow cytometry (FCM) was used to detect the expression of Survivin in 58 cases of colorectal carcinoma (CC) and 10 cases of normal mucosa (NM). SPSS11 was used in combination with clinical and pathological factors. 0 statistical software for statistical analysis. Results: FCM quantitative analysis showed that the average FI value of Survivin expression in normal mucosa group was 1.00 ± 0.28, and the FI values of Survivin expression in high, moderate and poorly differentiated adenocarcinoma and mucinous adenocarcinoma were 3.16 ± 1.39,3.20 ± 1.04,3.32 ± 0.48 And 3.31 ± 1.06. The expression of Survivin in high, moderate, poorly differentiated adenocarcinoma and mucinous adenocarcinoma were significantly higher than that in normal mucosa (P <0.05). The mean FI value of lymph node metastasis in colorectal cancer was 3.11 ± 1.19, which was significantly higher than that in non-lymph node metastasis (2.26 ± 1.38, P <0.05). The results of FCM showed that the expression of Survivin was not related to the patient’s age, sex, tumor location, etc. (P> 0.05). Conclusion: The expression of Survivin in colorectal cancer is abnormally elevated, which is significantly higher than that in normal intestinal mucosa. Survivin expression in colorectal cancer is closely related to lymph node metastasis.
