目的探讨扶正解毒颗粒(FJK)对镍诱导大鼠睾丸生精细胞凋亡的拮抗作用。方法选择健康成年Wistar雄性大鼠,NiSO42.5 mg/kg腹腔注射染毒,FJK 2.5、5.0和10.0 g/kg灌胃处理。30 d后,处死大鼠摘取睾丸,采用原位缺口末端标记法(TUNEL)检测生精细胞凋亡,免疫组化检测Bcl-2、Bax蛋白的表达水平。结果NiSO4染毒后,生精细胞总凋亡率升高(P<0.05),以生精周期Ⅰ~Ⅺ期、Ⅻ~ⅩⅢ期和ⅩⅣ期的精原细胞和精母细胞为主(P<0.05);精原细胞Bcl-2蛋白表达阳性细胞数减少,主要见于Ⅰ~Ⅵ期、Ⅻ~ⅩⅢ期和ⅩⅣ期(P<0.05),精母细胞为Ⅰ~Ⅵ期、Ⅻ~ⅩⅢ期多见(P<0.05);Bax蛋白表达阳性细胞数增多,精原、精母细胞均以Ⅰ~Ⅵ期、Ⅻ~ⅩⅢ期为主(P<0.05)。FJK处理后,上述异常变化有所改善,以10.0和5.0 g/kg为主(P<0.05)。结论FJK对NiSO4所致大鼠睾丸生精细胞凋亡具有拮抗作用,其机制可能与其引起Bcl-2表达上调和Bax表达下调有关。
Objective To investigate the antagonistic effect of Fuzheng Jiedu Granules (FJK) on nickel-induced apoptosis of rat spermatogenic cells. Methods Healthy adult male Wistar rats were selected, NiSO42.5 mg/kg was intraperitoneally injected, and FJK 2.5, 5.0, and 10.0 g/kg were intragastrically administered. After 30 days, the rats were killed and the testes were removed. The apoptosis of spermatogenic cells was detected by TUNEL. The expression of Bcl-2 and Bax protein was detected by immunohistochemistry. Results The total apoptosis rate of spermatogenic cells increased after NiSO4 exposure (P<0.05). The spermatogonia and spermatocytes of spermatogenic cycle I-XI, XII-XIII and XIV were predominant (P< 0.05); Spermatogonia Bcl-2 protein positive cells decreased, mainly seen in the I ~ VI period, XII ~ XIII and XIV period (P <0.05), spermatocytes for the I ~ VI period, XII ~ X III and more See (P<0.05); The number of Bax protein-positive cells increased, and spermatogonia and spermatocytes were mainly I-VI and XII-XIII (P<0.05). After the FJK treatment, the above abnormal changes were improved, with 10.0 and 5.0 g/kg predominating (P<0.05). Conclusion FJK can antagonize the apoptosis of spermatogenic cells in rat testis induced by NiSO4. Its mechanism may be related to its up-regulation of Bcl-2 expression and down-regulation of Bax expression.
