以巨噬细胞样细胞株,探讨了黄柏中对NO、细胞因子生成具有调节活性的成分。实验及结果:黄柏(230 g)用热水提取,浓缩后以3倍体积的乙醇使之沉淀,得粗多糖组分。粗多糖组分以冷冻溶解法分离,冷水不溶部分上DEAE Sephadex A-25(碳酸型)柱层析,经水洗脱后得中性糖组分。再以0.5 M(NH_4)2CO_3洗脱,得酸性糖组分。中性糖组分经Sephadex LH-20柱层析
In a macrophage-like cell line, components that have regulatory activity for NO and cytokine production in Phellodendron were investigated. Experiments and Results: Phellodendron (230 g) was extracted with hot water, concentrated and then precipitated with 3 volumes of ethanol to obtain crude polysaccharide fraction. The crude polysaccharide fraction was isolated by freeze-dissolving, and the cold water-insoluble fraction was subjected to column chromatography on DEAE Sephadex A-25 (carbonic acid type) and eluted with water to give a neutral sugar fraction. Elution with 0.5 M (NH_4)2CO_3 gave an acidic sugar fraction. Neutral sugar fractions by Sephadex LH-20 column chromatography
