目的探讨林可霉素对模型鼠脾细胞培养的上清液细胞因子白细胞介素17(IL-17)、IL-6和干扰素γ(IFN-γ)含量影响。方法以鸡Ⅱ型胶原(CCⅡ)和完全弗氏佐剂免疫DBA/1小鼠,建立小鼠胶原诱导性关节炎(CIA)动物模型。设正常对照组、CIA动物模型组和林可霉素CIA动物模型组,每组各6只。收集脾细胞培养的上清液,采用ELISA测定IL-17、IL-6及IFN-γ含量。结果林可霉素CIA动物模型组的IL-17、IL-6及IFN-γ含量均明显低于CIA动物模型组[(21.1±6.9)pg/ml vs.(33.4±10.79)pg/ml,(15.1±2.0)pg/ml vs.(22.5±5.8)pg/ml及(43.6±4.4)pg/ml vs.(59.3±16.1)pg/ml](P<0.05)。结论林可霉素能够抑制CCⅡ诱导小鼠关节炎的免疫反应。
Objective To investigate the effect of lincomycin on the contents of cytokines interleukin-17, interleukin-6 and interferon-γ in supernatant of splenocytes cultured in vitro. Methods The mouse model of collagen induced arthritis (CIA) was established by immunizing DBA / 1 mice with chicken type Ⅱ collagen (CC Ⅱ) and complete Freund’s adjuvant. The normal control group, CIA animal model group and lincomycin CIA animal model group, each group of 6. Supernatants from spleen cells were harvested and assayed for IL-17, IL-6 and IFN-γ by ELISA. Results The levels of IL-17, IL-6 and IFN-γin the CIA animal model group were significantly lower than those in the CIA animal model [(21.1 ± 6.9) pg / ml vs. (33.4 ± 10.79) pg / (15.1 ± 2.0) pg / ml vs. (22.5 ± 5.8) pg / ml and (43.6 ± 4.4) pg / ml vs. (59.3 ± 16.1) pg / ml], respectively. Conclusion Lincomycin can inhibit the immune response induced by CC Ⅱ in mice.
