T细胞需两种以上刺激信号才能充分活化。环孢素A(CsA)通过阻断CD3活化通路而产生免疫抑制效应,本文研究CsA对CD28活化通路的影响。研究发现以抗CD3单抗和抗CD28单抗共刺激后,淋巴细胞增殖比以抗CD3单抗单独刺激时显著增强,且对CsA不敏感,ID50较单独刺激时高4~5倍。研究表明:CD28通路能提供共刺激信号,使T细胞充分活化增殖,并且这一活化通路能对CsA产生抵抗
T cells need more than two stimulation signals to fully activate. Cyclosporine A (CsA) blocks the CD3 activation pathway to produce immunosuppressive effects. This article studies the effect of CsA on the CD28 activation pathway. The results showed that when stimulated with anti-CD3 monoclonal antibody and anti-CD28 monoclonal antibody, the lymphocyte proliferation ratio was significantly increased when stimulated with anti-CD3 monoclonal antibody alone, and was not sensitive to CsA. ID50 was 4 to 5 times higher than that when stimulated alone. Studies have shown that: CD28 pathway can provide costimulatory signals, T cells to fully activate and proliferate, and this activation pathway can be resistant to CsA
