目的探索慢性肺动脉高压大鼠心脏结构和功能的变化。方法雄性成年SD大鼠40只,肺动脉高压组32只,采用腹腔注射野百合碱溶液建立慢性肺动脉高压模型;对照组8只,注射同样剂量的乙醇和生理盐水混合液。注射28d后通过右心导管测定肺动脉压力,评价造模效果。肺动脉高压组大鼠于建模前后行超声检测,测量反映左右心室结构和功能的指标。结果成功建立慢性肺动脉高压模型大鼠29只。肺动脉高压组肺动脉收缩压、肺动脉平均压和右室收缩压均升高,与对照组比较差异有统计学意义(P<0.01)。超声测量结果显示肺动脉高压组大鼠干预后舒张末期右心室上下径显著增大(P<0.05),左心室舒张末期横径及上下径变化不显著(P>0.05)。二尖瓣、三尖瓣E/A明显降低,与干预前相比差异有统计学意义(P<0.05)。左室射血分值、短轴缩短率及每搏量干预前后差异不具有显著性(P>0.05)。结论慢性肺动脉高压大鼠右室结构改变,左右心室的舒张功能下降,短期内左室收缩功能未发生明显改变。
Objective To explore the changes of cardiac structure and function in chronic pulmonary hypertension rats. Methods Forty adult male Sprague-Dawley rats and 32 pulmonary hypertension rats were randomly divided into four groups: control group (n = 12) and control group (n = 10). The control group was injected with the same dose of ethanol and saline solution. Pulmonary artery pressure was measured by right heart catheter 28 days after injection, and the modeling effect was evaluated. Pulmonary hypertension rats before and after modeling ultrasound detection, measurement of left and right ventricular structural and functional indicators. Results 29 rats with chronic pulmonary hypertension were successfully established. The pulmonary arterial systolic pressure, pulmonary arterial pressure and right ventricular systolic pressure in pulmonary arterial hypertension group were significantly higher than those in control group (P <0.01). The results of ultrasound showed that the diameter of the right ventricle and the diameter of the left ventricle in the pulmonary hypertension group were significantly increased after intervention (P <0.05), while the changes of the diameter and the diameter of the left ventricular end diastolic were not significant (P> 0.05). Mitral, tricuspid valve E / A was significantly lower than before intervention, the difference was statistically significant (P <0.05). Left ventricular ejection fraction, short axis shortening rate and stroke volume before and after intervention did not differ significantly (P> 0.05). Conclusion Right ventricular structure changes in rats with chronic pulmonary hypertension, left and right ventricular diastolic function decreased, short-term left ventricular systolic function did not change significantly.
