目的 :研究 p5 3基因点突变在食管癌及癌前病变粘膜细胞癌变过程中的变异作用及定量分析 ,对病人预后的关系。方法 :选取手术新鲜标本及蜡块 90例。DNA提取分析 ,应用PCR -RFLP和DNA序列分析 ,免疫组化ABC法 ,利用MPV - 2型显微分光光度计检测分析。结果 p5 3基因在食管癌、不典型增生组织中点突变频率较高 ,分别为 6 0 %、2 9.4%。错义突变和移码突变占全部突变的 73%。p5 3表达强度与生存期有相关性。结论 :p5 3基因点突变在食管粘膜细胞癌变过程中起重要作用。
Objective: To study the variation and quantitative analysis of point mutation of p5 3 gene in the carcinogenesis of esophageal cancer and precancerous mucosa, and its relationship with the prognosis of patients. Methods: Fresh specimens and 90 cases of wax block were selected. DNA extraction and analysis, PCR - RFLP and DNA sequence analysis, immunohistochemical ABC method, using MPV - 2 spectrophotometer detection analysis. Results The frequency of point mutation of p5 3 gene in esophageal cancer and atypical hyperplasia was 60% and 294% respectively. Missense mutations and frameshift mutations accounted for 73% of all mutations. p5 3 expression intensity and survival were related. Conclusion: The point mutation of p5 3 gene plays an important role in the carcinogenesis of esophageal mucosa.
