目的检测妊娠不同阶段正常孕妇外周血单个核细胞(PBMC)IL-8基因表达水平及血清中IL-8含量。方法用密度梯度离心法分离正常孕妇PBMC,调整细胞浓度为1×106,提取RNA,逆转录合成cDNA,用荧光定量PCR方法检测IL-8mRNA表达水平。同时取正常孕妇外周血,分离血清,用ELISA和化学发光法检测孕妇血清中IL-8及人绒毛膜促性腺激素(HCG)含量。结果在妊娠初期,IL-8的表达(0.029±0.010)和分泌([49.42±6.68)ng/ml]随着HCG含量的增加达到最高,在妊娠中期和晚期,IL-8的表达(中期0.004±0.001,晚期0.005±0.001)和分泌[中期(8.17±3.09)ng/ml,晚期(7.98±3.05)ng/ml]又降到较低的水平。结论 IL-8在妊娠初期的表达水平明显高于妊娠中期和晚期,提示,在HCG作用下IL-8参与了妊娠的启动。
Objective To detect the expression of IL-8 in peripheral blood mononuclear cells (PBMCs) and IL-8 levels in serum of pregnant women at different stages of pregnancy. Methods PBMCs from normal pregnant women were isolated by density gradient centrifugation. The concentration of RNA was adjusted to 1 × 106. RNA was extracted and cDNA was reverse transcribed. The expression of IL-8 mRNA was detected by real-time PCR. At the same time, the peripheral blood of normal pregnant women was taken out and the serum was separated. The levels of IL-8 and human chorionic gonadotropin (HCG) in pregnant women were detected by ELISA and chemiluminescence. Results In the first trimester of pregnancy, the expression of IL-8 (0.029 ± 0.010) and secretion ([49.42 ± 6.68] ng / ml] reached the peak with the increase of HCG content. The expression of IL-8 in the second trimester of pregnancy ± 0.001, late 0.005 ± 0.001) and secretion [interim (8.17 ± 3.09) ng / ml, late (7.98 ± 3.05) ng / ml] Conclusion The expression of IL-8 in early pregnancy is obviously higher than that in middle and late pregnancy, suggesting that IL-8 is involved in the initiation of pregnancy under the action of HCG.
