目的 :研究山茱萸补益活性成分。方法 :采用热水提取 ,乙醇沉淀 ,三氯醋酸除蛋白等步骤分得粗多糖 ,DEAE 纤维素及SephadexG 2 0 0柱层析分离 ,纯化 ,高效凝胶渗透色谱鉴定纯度并测定分子量 ,薄层层析 ,纸层析 ,气相色谱 ,气 质联用 ,甲基化分析 ,部分酸水解及13CNMR确定多糖的化学结构。结果 :分离得到一水溶性多糖Co 4 ,其分子量为 2 .4 7× 1 0 4 ,主链为 1 ,4 连接的 β D 葡聚糖 ,部分葡萄糖残基 6位上有分枝。结论 :首次对山茱萸多糖进行较深入的化学研究并阐明水溶性多糖Co 4化学结构。
Objective : To study the active ingredients of Hawthorn replenishing. METHODS: The crude polysaccharides were extracted by hot water extraction, ethanol precipitation, trichloroacetic acid and protein removal, DEAE cellulose and Sephadex G200 column chromatography, purification, and high performance gel permeation chromatography to determine the purity and determine the molecular weight. Chromatography, paper chromatography, gas chromatography, GC-MS, methylation analysis, partial acid hydrolysis and 13C NMR were used to determine the chemical structure of polysaccharides. RESULTS: A water-soluble polysaccharide Co 4 was isolated. Its molecular weight was 2.47 × 10 4 , the main chain was 1, 4 linked β D glucan, and some glucose residues were branched on the 6th position. Conclusion : For the first time, the polysaccharides of Hawthorn were studied in depth and the chemical structure of water-soluble polysaccharide Co 4 was elucidated.
