目的:研究清脑宣窍滴丸对急性脑缺血/灌注损伤引起的一氧化氮(NO)含量和一氧化氮合酶(NOS)活力的改变及机制。方法:采用线栓法致大鼠大脑中动脉缺血再灌注(MCAO)模型,测定大鼠手术侧脑匀浆中NO含量及结构型一氧化氮合酶(cNOS)、诱导型一氧化氮合酶(iNOS)、总NOS活力。结果:清脑宣窍滴丸45,90,180 mg.kg-1可明显降低MCAO大鼠患侧脑匀浆中NO含量,180 mg.kg-1可明显降低术侧大脑组织升高的总NOS,cNOS和iNOS活力(P<0.05);90 mg.kg-1也可明显降低升高的总NOS活力(P<0.01)。45 mg.kg-1明显降低升高的总NOS活力(P<0.05)。结论:清脑宣窍滴丸可能通过减少NO含量,降低iNOS的活性来防治脑缺血/再灌注损伤。
Objective: To study the changes of nitric oxide (NO) content and nitric oxide synthase (NOS) activity induced by Qingnao Xuanqi Dripping Pill in acute cerebral ischemia/reperfusion injury. METHODS: The rat middle cerebral artery occlusion (MCAO) model was induced by suture embolization. The content of NO, cNOS and inducible nitric oxide were measured in the rat brain after surgery. Enzyme (iNOS), total NOS activity. RESULTS: Qingnao Xuanqi Dripping Pills 45, 90, 180 mg.kg-1 significantly reduced the content of NO in the homogenate of the affected side of MCAO rats, and 180 mg.kg-1 significantly decreased the total NOS in the lateral cerebral tissue. The activity of cNOS and iNOS (P<0.05); 90 mg.kg-1 also significantly decreased the total NOS activity (P<0.01). 45 mg.kg-1 significantly reduced the elevated total NOS activity (P<0.05). Conclusion: Qingnao Xuanqi Dripping Pills may prevent cerebral ischemia/reperfusion injury by reducing the content of NO and decreasing the activity of iNOS.
