目的探讨疏血通注射液是否能影响IIa型分泌型磷脂酶A2(s PLA2 IIa)的表达并分析其可能的作用机制。方法通过建立大鼠动脉粥样硬化模型,观察s PLA2 IIa在食饵性动脉粥样硬化模型大鼠主动脉的表达。30只Wistar大鼠,随机分为正常对照组、高脂模型组、疏血通组3组(n=10)。对照组饲以普通饲料,高脂模型组大鼠给予高脂饲料喂养,疏血通组大鼠给予高脂饲料喂养并予疏血通注射液[4 m L/(kg·d)]静脉注射,持续12周。监测血糖、血压;免疫组织化学法及western blot法检测3组大鼠主动脉中s PLA2 IIa的表达。结果在高脂模型组、疏血通组大鼠主动脉血管平滑肌与动脉粥样硬化斑块中明显可见s PLA2 IIa的表达。高脂模型组大鼠的主动脉壁中s PLA2 IIa表达较正常对照组明显增加(P<0.01);疏血通组大鼠主动脉壁的s PLA2 IIa表达与高脂模型组相比,明显减少(P<0.01)。结论 s PLA2 IIa表达的增加可能是导致动脉粥样硬化的重要原因之一,疏血通注射液可抑制s PLA2 IIa在主动脉的表达,从而减轻动脉粥样硬化。
Objective To investigate whether Shuxuetong injection can affect the expression of type IIa sPL2a and its possible mechanism. Methods The rat model of atherosclerosis was established to observe the expression of s PLA2 IIa in the aorta of rats with preeclamptic atherosclerosis. 30 Wistar rats were randomly divided into normal control group, high fat model group and Shuxuetong group (n = 10). The rats in the control group were fed with normal diet, and the rats in the high fat model group were fed with high fat diet. The rats in Shuxuetong group were fed with high fat diet and given Shuxuetong injection [4 m L / (kg · d)] intravenously For 12 weeks. The blood glucose and blood pressure were monitored. The expression of PLA2IIa in the aorta was detected by immunohistochemistry and western blot. Results The expression of sPLA2IIa in the aortic smooth muscle and atherosclerotic plaque of the Hyperlipidemic model group and Shuxuetong group was obvious. Compared with the normal control group, the expression of PLA2IIa in the aortic wall of rats in the high-fat model group was significantly increased (P <0.01). The PLA2IIa expression in the aorta of Shuxuetong group was significantly higher than that in the model group Decrease (P <0.01). Conclusion The increase of PLA2IIa expression may be one of the most important causes of atherosclerosis. Shuxuetong injection can inhibit the expression of sPLA2IIa in the aorta and relieve atherosclerosis.
