采用RT-PCR方法从‘紫罗兰’牡丹(Paeonia suffruticosa‘Ziluolan’)花芽中克隆得到1个重要开花调控转录因子基因CONSTANS-like,其cDNA全长1 125 bp,编码373个氨基酸。序列比对和结构域分析表明,此蛋白包含两个B-box型锌指结构和1个CCT结构域,与拟南芥的AtCOL4最为相似,将其命名为PsCOL4,GenBank登录号为KF113358。系统进化树分析表明,PsCOL4与葡萄编码的VvCOL4的亲缘关系最近,属于第1群组CO类似基因。实时定量PCR表明,PsCOL4在不同组织器官中均有表达,其中茎和叶中的表达量最高,根中最低。在花芽不同发育时期,PsCOL4的表达呈现递减趋势。不同光周期条件下,PsCOL4的表达稍有不同,短日照条件下在叶中的表达有所增加,茎中稍有减少。
One important flowering regulatory transcription factor gene, CONSTANS-like, was cloned by RT-PCR from the flower buds of Paeonia suffruticosa’Ziluolan ’. The full length cDNA was 1 125 bp and encoded 373 amino acids. Sequence alignment and domain analysis indicated that this protein contains two B-box type zinc finger structures and one CCT domain, which is the most similar to AtCOL4 in Arabidopsis thaliana, and named PsCOL4 as GenBank accession number KF113358. Phylogenetic tree analysis showed that the closest relationship between PsCOL4 and VvCOL4 encoding grape belongs to the first group of CO-like genes. Real-time quantitative PCR showed that PsCOL4 was expressed in different tissues and organs, of which the highest expression was found in stem and leaf and the lowest in root. The expression of PsCOL4 showed a decreasing trend at different developmental stages of flower buds. The expression of PsCOL4 was slightly different under different photoperiod conditions. The expression of PsCOL4 was slightly increased in short-day conditions and slightly in stems.
