为了促进SNP技术在番茄抗黄化曲叶病毒病育种中的应用,探讨了将番茄Ty-5 CAPS标记转换成SNP标记进行检测的可行性。首先对Ty-5的CAPS标记Sl NAC1的扩增产物进行测序,设计多对引物,以感病自交系9179和抗病自交系07-025为材料进行PCR,筛选出只在抗病自交系中能扩增出特异条带的SNP引物,将这对引物作为SNP标记;然后,用SNP标记对番茄材料进行分析,验证标记的可行性。结果表明,利用番茄Ty-5 CAPS标记转化的SNP标记,可直接用于番茄抗黄化曲叶病毒病的分子标记辅助选择育种。
In order to promote the application of SNP technology in the breeding of tomato leaf yellow curl virus, the feasibility of detecting the tomato Ty-5 CAPS marker converted to SNP marker was discussed. Firstly, the amplification products of CA N-labeled S1 NAC1 of Ty-5 were sequenced, and several pairs of primers were designed. The susceptible inbred lines 9179 and the resistant inbred lines 07-025 were selected for PCR. The SNP primers that can amplify specific bands in the cross can be used as the SNP markers. Then, the SNP markers were used to analyze the tomato materials to verify the feasibility of the markers. The results showed that the SNP marker transformed by tomato Ty-5 CAPS marker could be directly used for molecular marker-assisted selection breeding of tomato leaf yellow curl virus disease.
