目的探讨补肾化瘀方对载脂蛋白E基因缺陷(ApoE-/-)小鼠头臂干动脉粥样硬化斑块稳定性和斑块内细胞凋亡水平的影响及可能机制。方法 24只8周龄雄性ApoE-/-小鼠饲以“西方类型”膳食(高脂饲料)20周后,头臂干已经形成易损动脉粥样硬化(AS)斑块,随机将小鼠分为3组:补肾化瘀组、他汀组和盐水组,连续给药12周后处死动物,将头臂干动脉连续石蜡切片、HE染色后,观察小鼠动脉粥样斑块的形态,并用TUNEL法检测斑块内细胞凋亡水平。结果补肾化瘀方干预后动脉粥样斑块的校正脂质核面积(LCA/PA)减小,斑块纤维帽增厚,斑块趋于稳定;补肾化瘀组斑块中细胞凋亡指数(AI)显著降低(P<0.05),斑块内Bcl-2蛋白表达升高、Bax蛋白表达显著降低(P<0.01)。结论补肾化瘀方可不同程度地减小脂质核面积,增加纤维帽厚度,抑制斑块内细胞凋亡,起到稳定AS斑块的作用。
Objective To investigate the effects of Bushen Huayu Recipe on the stability of atherosclerotic plaque in the brachial trunk of ApoE - / - deficient mice and the possible mechanism. Methods Twenty-four male ApoE - / - mice, aged 8 weeks, were given “Western type” diet (high fat diet) for 20 weeks and their brachial trunk had formed vulnerable atherosclerotic plaque. The mice were divided into three groups: Bushen Huayu group, statin group and saline group. After continuous administration for 12 weeks, the animals were sacrificed and the paraffin sections of the trunk arteries were continuously paraffinized. The morphology of the atherosclerotic plaque was observed after HE staining , And TUNEL method was used to detect the level of apoptosis in plaque. Results After the intervention of Bushen Huayu Recipe, the corrected LCA / PA of atherosclerotic plaque was reduced, the plaque cap was thicker and the plaque tended to be stable. The apoptosis index (AI) (P <0.05). The expression of Bcl-2 protein and Bax protein in plaque were significantly decreased (P <0.01). Conclusion Bushen Huayu Recipe can reduce the area of lipid nucleus to a certain extent, increase the thickness of fiber cap, inhibit the apoptosis in plaque and play a role in stabilizing AS plaque.
