裸鼠净化饲养室的建立使用与管理

为能自行提供生存期较长的健康裸鼠用于裸鼠人体肝癌模型的研究,我室于1983年5月建立了顶送下侧回风紊流型裸鼠净化饲养室。建筑面积为54平方米,净化饲养面积20平方米。经测试,饲养室净化级别为1000级,温湿度分别控制在24~26℃和40~60%范围,换气次数为10~15次/小时,裸鼠生活区的风速为0.1米/秒,均达原设计要求。经一年多的试用,各机组运转基本正常,裸鼠发育、繁殖良好,生存期明显延长,长期观察的裸鼠一年存活率达80%。裸鼠人体肝癌移植模型已经建立,并用于肝癌肿瘤标记、阳性扫描及实验性治疗等研究。以动物为宿主建立人癌模型一直引起人们的兴趣并为广大学者所关注。但往往因免疫排斥反应而难以获得成功。只是当无胸腺裸鼠出现后,这个难题才得以解决。显而易见,裸鼠的能否提供及其质量直接影响该项科研的成败。然而,裸鼠因免疫功能低下难于饲养和繁殖,因此,建立裸鼠所需的无特定病原菌(SPF)环境,保征裸鼠长期存潘已成为进行裸鼠人癌研究的前提。我室为开展裸鼠人体肝癌移植模型的研究,于1979年底引入裸小鼠,饲养于无严格空气过滤的课鼠室,虽裸鼠尚能存活,人肝癌异种移植也可成功,但正如文献所述,在常规环境裸鼠寿命较短,易患肝炎,无法满足长期实验的要求。为此,决定建立裸鼠净化饲养室自行培育较高质量的裸小鼠。经10个月的施工及调试,该室于1983年5月正式使用。经一年多的试用,各机组运转基本正常,各项指标符合设计要求。目前,裸小鼠发育繁殖良好,生存期明显延长。长期观察的裸鼠,一年存活率达80%。裸鼠人体肝癌,模型已经建立并用于肝癌的基础和临床研究。本文报告裸鼠净化饲养室的有关情况及使用体会。 In order to provide healthy nude mice with long-lived life for the study of nude mice’s human liver cancer models, our laboratory established a scavenging feeding room for top-feeding lower air return turbulence nude mice in May 1983. The building area is 54 square meters and the purifying breeding area is 20 square meters. After testing, the breeding room has a purification level of 1000, and the temperature and humidity are controlled at 24 to 26°C and 40 to 60% respectively, the number of ventilations is 10 to 15 times/hour, and the nude mouse’s living area has a wind speed of 0.1 m/s. All reached the original design requirements. After more than one year’s trial, the units were running normally, and the nude mice developed well and had a good reproduction. The survival period was prolonged. The long-term observed nude mice had a survival rate of 80%. The human hepatoma transplant model in nude mice has been established and used for studies of tumor markers, positive scans, and experimental treatments for liver cancer. The establishment of a human cancer model with animals as the host has always attracted people’s interest and is of great interest to the majority of scholars. However, it is often difficult to achieve success due to immune rejection. It was only when the athymic nude mouse appeared that this problem was solved. Obviously, the availability and quality of nude mice directly affects the success or failure of the research. However, it is difficult for nude mice to breed and reproduce due to low immune function. Therefore, the establishment of a specific pathogen-free (SPF) environment for nude mice to protect long-term nude mice has become a prerequisite for the study of human cancer in nude mice. Our laboratory developed a nude mouse model of human liver cancer transplantation. At the end of 1979, nude mice were introduced and raised in a classroom without strict air filtration. Although nude mice survived, human liver cancer xenotransplantation was successful, but as the literature As mentioned above, in a normal environment, nude mice have a short life span and are prone to hepatitis, and cannot meet the requirements of long-term experiments. For this reason, it was decided to establish a nude mice breeding room to cultivate nude mice of higher quality on their own. After 10 months of construction and commissioning, the room was formally used in May 1983. After more than one year of trials, the units were operating normally and all the indicators met the design requirements. At present, nude mice develop well and have a prolonged survival period. Long-term observation of nude mice, one year survival rate of 80%. Human liver cancer in nude mice has been established and used for basic and clinical studies of liver cancer. This article reports on the status and use experience of a nude mouse clean room.
