目的 评估逆转录聚合酶链反应 (reversetranscriptasepolymerasechainreaction ,RT PCR)、免疫组化 (immunohistochemistry ,IHC)和微粒子酶免疫测定法 (microparticalenzymeimmunoassay ,MEIA)检测裸大鼠体内人结肠癌 (HCC)细胞的敏感性和特异性。方法 应用HCC细胞系HT 2 9建立裸大鼠HCC转移模型 ,通过RT PCR检测细胞角蛋白 2 0 (cytokeratin 2 0 ,CK 2 0 )mRNA的表达、免疫组化APAAP法检测人CK、MEIA测定组织癌胚抗原 (carcinoembryonicantigen ,CEA)和常规病理学 (routinepathology,RP)等方法对裸鼠肝肺组织中HCC细胞进行检测。结果 RT PCR、IHC、MEIA和RP的HCC细胞阳性检出率分别为 73 8%、64 3 %、33 %和 50 %。RT PCR阳性率最高 ,与RP和MEIA相比 ,差异均有显著性意义 (χ2 =5 0 4 8,P <0 0 5和 χ2 =1 3 832 ,P <0 0 0 1 )。IHC阳性率与MEIA结果比较 ,差异也有显著性意义 (χ2 =8 0 52 ,P <0 0 1 )。结论 应用RT PCR检测CK2 0mRNA表达和APAAP法检测人CK表达在裸大鼠HCC转移模型内检测癌细胞具有高敏感性和高特异性。MEIA测定组织CEA具有高特异性和低敏感性
Objective To evaluate the sensitivity and specificity of reverse transcriptase polymerase chain reaction (RT PCR), immunohistochemistry (IHC) and microparticulate enzyme immunoassay (MEIA) in detecting nude rat colon carcinoma (HCC) Specificity. Methods HCC cell line HT 2 9 was used to establish the model of HCC metastasis in nude mice. The expression of cytokeratin 2 0 (CK 2 0) mRNA was detected by RT PCR. The expression of CK 2 0 was examined by immunohistochemical APAAP method. Carcinoembryonic antigen (CEA) and routine pathology (RP) were used to detect HCC cells in liver and lung tissue of nude mice. Results The positive rates of HCC cells in RT PCR, IHC, MEIA and RP were 73 8%, 64 3%, 33% and 50%, respectively. The positive rate of RT PCR was highest, which was significantly different from that of RP and MEIA (χ2 = 5084, P <0.05 and χ2 = 13832, P <0.01). IHC positive rate and MEIA results, the difference was also significant (χ2 = 8 0 52, P <0 0 1). Conclusion The detection of CK20 mRNA by RT-PCR and the detection of human CK expression by APAAP are highly sensitive and specific in the detection of cancer cells in nude rat model of HCC metastasis.
