目的:研究游离大黄蒽醌类化合物对骨质疏松模型大鼠骨密度(BMD)、骨矿含量(BMC)的影响。方法:灌胃醋酸泼尼松(3.5 mg/kg),每日1次,以复制大鼠骨质疏松模型。4月龄126只雌性SD大鼠随机均分为正常对照(等容生理盐水)组、模型(等容生理盐水)组、游离大黄蒽醌(70 mg/kg)组,每组再分1、3、5、7、9、11、13周7个时间点。上午灌胃游离大黄蒽醌,下午灌胃醋酸泼尼松。在相应时间点,大鼠麻醉后心脏抽血至尽处死,采集四肢骨,以pQCT骨密度仪分别扫描大鼠股骨全骨和股骨远端(2 mm),测定BMD和BMC。结果:与正常对照组比较,复制模型1周后,模型组大鼠股骨远端BMD显著减小;复制模型3周后,模型组大鼠股骨全骨BMD显著减小;复制模型5周后,模型组大鼠股骨远端BMC显著减少(P<0.05)。与模型组比较,在7个时间点上游离大黄蒽醌组大鼠股骨全骨和股骨远端的BMD显著增加(P<0.05)。结论:游离大黄蒽醌可预防醋酸泼尼松所致的股骨BMD的丢失,未观察到大鼠股骨BMC随时间出现显著蓄积的现象。
Objective: To study the effects of free rhubarb anthraquinones on bone mineral density (BMD) and bone mineral content (BMC) in rats with osteoporosis. Methods: Prednisone acetate (3.5 mg / kg) was administered orally once a day to replicate the rat model of osteoporosis. One hundred and sixty-four female SD rats at 4 months of age were randomly divided into normal control (isoboric saline) group, model group (isocitrate saline) and free rhubarb anthraquinone (70 mg / kg) 3,5,7,9,11,13 weeks 7 time points. Free rhubarb anthraquinone gavage in the morning and prednisone acetate in the afternoon. At the corresponding time point, the hearts of the rats were drawn to death after anesthesia. The extremities bone were collected and the total femur and femur distal ends (2 mm) were scanned with pQCT bone densitometer. The BMD and BMC were measured. Results: Compared with the normal control group, the BMD of the distal femur in model group decreased significantly after 1 week of replication. After 3 weeks of replication model, the BMD of femoral total bone in model group decreased significantly. After 5 weeks of replication model, The BMC in distal femur of the model group was significantly decreased (P <0.05). Compared with the model group, the BMD of the femur and the distal femur of rats in the rhubarb-anthraquinone group was significantly increased at 7 time points (P <0.05). CONCLUSION: Free rhubarb anthraquinone can prevent the loss of femoral BMD caused by prednisone acetate. No significant accumulation of BMC in rat femur was observed over time.
