利用免疫组化技术研究ATX在胃癌及正常人胃粘膜组织之间表达的差异,Western Blot法检测ATX表达及ERK磷酸化,MTT法测定细胞增殖,Boyden chamber法测定细胞迁移.结果显示:ATX在正常胃粘膜组织、癌旁组织及胃癌组织均有表达,胃癌细胞BGC-803中有表达;ATX具有溶血磷脂酶D活性,能将LPC水解为LPA,并以LPA受体介导信号通路促进胃癌细胞的增殖和迁移作用.由此得出LPC是通过LPC-Autotaxin-LPA-LPA1/3-Gi-ERK信号通路诱导胃癌细胞增殖与迁移.ATX/LPA通路在胃癌治疗中也是一个潜在的靶位点.
The expression of ATX in gastric cancer and normal gastric mucosa was studied by immunohistochemistry, the ATX expression and ERK phosphorylation were detected by Western Blot, the cell proliferation was measured by MTT assay and the cell migration was measured by Boyden chamber.The results showed that ATX Normal gastric mucosa tissues, paracancerous tissues and gastric cancer tissues were expressed in gastric cancer cells BGC-803; ATX with lysophospholipase D activity, hydrolysis of LPC to LPA, LPA receptor-mediated signaling pathway to promote gastric cancer LPC-Autotaxin-LPA1 / 3-Gi-ERK signaling pathway to induce the proliferation and migration of gastric cancer cells.ATX / LPA pathway is also a potential target in the treatment of gastric cancer point.
