探讨了碱性成纤维细胞生长因子(basicfibroblastgrowthfactor,bFGF)能否对人结膜下Tenon’s囊成纤维细胞(humansubconjunctivalTenon’scapsulefibroblast,HTCF)表型改变起诱导作用。在5例白内障手术中取人结膜下Tenon’s囊组织块培养成纤维细胞。用不同浓度bFGF(0、5、10、20ng/ml)诱导HTCF48h。免疫细胞化学技术、蛋白质印记免疫技术检测其平滑肌肌动蛋白(α-smoothmuscleactin,α-SMA)表达与正常HTCF有否差异。实验结果表明用不同浓度(0 ̄20ng/ml)bFGF诱导HTCF48h,虽能明显促进HTCF细胞的生长,但均不能上调细胞内α-SMA的表达。在0 ̄20ng/ml范围内,体外用bFGF诱导HTCF48h,不能诱导其改变为肌成纤维细胞。
The purpose of this study was to investigate whether basic fibroblast growth factor (bFGF) can induce phenotypic changes in human subconjunctival Tenon’s capsule fibroblasts (HTCF). Fibroblasts were cultured in subconjunctival Tenon’s sac in 5 cataract surgeries. HTCF was induced for 48 h with different concentrations of bFGF (0, 5, 10, 20 ng / ml). Immunocytochemistry and Western blotting were used to detect the expression of α-smooth muscle actin (α-SMA) and normal HTCF. The experimental results showed that induction of HTCF48h with different concentrations of bFGF (0 ~ 20ng / ml) could significantly promote the growth of HTCF cells, but neither up-regulated the expression of α-SMA. In the range of 0-20 ng / ml, HTCF48h was induced with bFGF in vitro and could not be induced to change into myofibroblasts.
