野菊花对实验性心室重构的影响

目的:研究野菊花水提液对异丙肾上腺素(ISO)诱发小鼠心室重构及L-甲状腺素(L-Thy)诱发大鼠实验性心室重构的影响。方法:小鼠每天scISO2mg/kg,连续7d;大鼠每天ipL-Thy0.25mg/kg,连续9d,制备心室重构动物模型。小鼠经药物干预7d后,称重法测定心脏指数(HW/BW)及左心室指数(LVW/BW);放射免疫法测定心肌血管紧张素Ⅱ(AngⅡ)含量;大鼠经药物干预9d后,称重法测定心脏指数(HW/BW)及左心室指数(LVW/BW);放射免疫法测定心肌血管紧张素Ⅱ(AngⅡ)、血清醛固酮(ALD)和肿瘤坏死因子(TNF-α)的水平;心肌样本碱水法测定羟脯氨酸(Hyp)的含量。结果:与正常对照组比较,给药7d时ISO诱发心室重构模型小鼠的HW/BW、LVW/BW及AngⅡ含量升高(P<0.05),野菊花水提液能显著改善模型小鼠心脏指数,降低心肌AngⅡ含量(P<0.05)。与正常对照组比较,给药9d时,L-Thy诱发心室重构模型大鼠HW/BW、LVW/BW,AngⅡ、ALD、TNF-α及Hyp含量均显著升高(P<0.05),野菊花水提液能显著改善大鼠心脏指数,降低血清ALD、TNF-α和心肌AngⅡ、Hyp水平(P<0.05)。结论:野菊花具有抑制实验动物心室重构的作用,其作用机制可能与抑制交感神经系统过度兴奋,降低AngⅡ、ALD和TNF-α等神经内分泌因子过度表达有关。 Objective: To study the effect of wild chrysanthemum extract on isoproterenol (ISO) -induced ventricular remodeling and L-Thy-induced experimental ventricular remodeling in rats. CONCLUSION: The wild chrysanthemum can inhibit the ventricular remodeling in experimental animals. Its mechanism may be related to inhibiting the over-excitement of the sympathetic nervous system and decreasing the expression of neuroendocrine factors such as AngⅡ, ALD and TNF-α.
