目的:完善和提高白背叶根的质量标准。方法:显微鉴别白背叶根横切面和粉末;采用TLC法对白背叶根水溶性成分和脂溶性成分进行定性鉴别;特征图谱采用HPLC法以芦丁为参照物确定各特征峰的相对保留时间及相对峰面积,色谱柱:SPOLAR C18柱(250 mm×4.6 mm,5μm),流动相:乙腈-0.4%磷酸溶液梯度洗脱;检测波长为328 nm;流速:1.0 ml·min-1;柱温:35℃。结果:显微特征明显;薄层色谱斑点圆整、清晰、重复性好。特征图谱供试品色谱呈现5个特征峰,与参照物(芦丁)峰的相对保留时间应在规定值±5%之内,其中峰3、峰4相对峰面积不低于0.23和0.66。结论:所建立的方法简便快捷,重复性好,结果准确可靠,能有效控制白背叶根质量,可作为白背叶根的质量标准的主要控制指标。
Objective: To improve and improve the quality standard of white dorsal root. Methods: The cross section and powder of white dorsal root were identified by microscopy. The water-soluble and fat-soluble components of white dorsal radish were identified qualitatively by TLC. The relative retention of each peak was determined by HPLC with rutin as reference Time and relative peak area. The chromatographic column was SPOLAR C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase was eluted with acetonitrile-0.4% phosphoric acid solution. The detection wavelength was 328 nm. The flow rate was 1.0 ml · min -1. Column temperature: 35 ° C. Results: The microscopic features were obvious. The TLC spots were round and clear with good repeatability. The characteristic chromatogram showed five characteristic peaks for the chromatogram of the test product. The relative retention time with the reference (rutin) peak should be within ± 5% of the specified value, and the relative peak area of peak 3 and peak 4 should not be less than 0.23 and 0.66. Conclusion: The established method is simple, rapid and reproducible, the results are accurate and reliable, can effectively control the quality of white dorsal root and can be used as the main control index of the quality standard of white dorsal root.
