酶法制备头孢氨苄的转化液中有一些与头孢氨苄结构相似的底物和前体等物质。为了从转化液中分析头孢氨苄的含量,我们设计并建立了用反相HPLC法分析测定头孢氨苄的效价,并及时反馈转化液、吸附废液、解析液、固体粗品中头孢氨苄的含量,为中间体各环节的质量控制提供了一个简便、快速、灵敏、准确的方法。实验采用Waters液相色谱仪,C18柱,流动相为甲醇、水、冰乙酸混合液,在2~60μg/ml浓度范围内线性关系良好,r=0.9999(n=6),平均回收率为99.59%,RSD为0.53%。
